In vivo study | GSK-2269557 has a relatively low oral bioavailability (F = 2%), with a clearance of 28 mL/min/kg, but a high volume distribution of 6.3 L/kg. 0.3-1 μMGSK-2269557 induced concentration-dependent prolongation of QT interval and T-p-e, and 1 μm GSK-2269557 resulted in prolongation of the ORS wave, during which no tip-torsion arrhythmia was observed. In an acute OVA disease model in brown Norway rats, Th2 drives lung inflammation and is GSK-2269557 able to protect the lungs from eosinophil recruitment (ED50=67 μg/kg), and concentration-dependently reduce recruitment and IL-13 of all leukocyte subpopulations. |
